Abstract: Bovine spongiform encephalopathy (BSE) is a fatal neurological
disease of cattle, recognized in Great Britain in 1986. Cases in other
countries have been attributed to imports from Great Britain. The
disease has not occurred in the U.S. BSE is one of a group of diseases
(other examples are scrapie of sheep and Creutzfeld-Jacob disease of
humans) referred to as prion diseases or transmissible spongiform
encephalopathies. Under some circumstances prion diseases can be
transmitted by injection or by feeding infected (abnormal prion
protein-containing) tissue to susceptible hosts. BSE was disseminated
by feeding meal and bone meal containing BSE agent which was not
completely inactivated by rendering, BSE is hypothesized to have
emerged from scrapie via recycling of rendered by-products in cattle.
There is also evidence of spontaneous feed-borne transmission of BSE to
wild ruminants in zoological parks and to domestic cats. It has been
hypothesized that foodborne transmission of BSE to humans has occurred
or could occur. This hypothesis can neither be definitively refuted nor
supported, However, it seems unlikely. In spite of hundreds of years of
human exposure to scrapie, there is no evidence of transmission of
scrapie to humans. Even if BSE is ultimately found to be somehow
transmissible to humans, the risk of foodborne transmission appears to
be low for several reasons: (i) The oral route is several orders of
magnitude less sensitive than the parenteral route for transmission of
prion diseases; (ii) the BSE agent is only detectable in brain, spinal
cord, and intestine of infected cattle, tissues infrequently used for
human food; and (iii) Great Britain (where the disease occurs) destroys
and bans the use of all tissues from BSE-infected cattle as well as the
brains, spinal cords, and intestinal tracts from clinically normal
cattle.

Abstract: Strains of transmissible spongiform encephalopathies are
distinguished by differing physicochemical properties of PrPSc, the
disease-related isoform of prion protein, which can be maintained on
transmission to transgenic mice. 'New variant' Creutzfeldt-Jakob
disease (CJD) has strain characteristics distinct from other types of
CJD and which resemble those of BSE transmitted to mice, domestic cat
and macaque, consistent with BSE being the source of this new disease.
Strain characteristics revealed here suggest that the prion protein may
itself encode disease phenotype.

Abstract: Bovine spongiform encephalopathy (BSE), also known as ''mad cow
disease,'' is a fatal brain disease of cattle first recognized in the
United Kingdom. In humans, the most common transmissible spongiform
encephalopathy is Creutzfeldt-Jacob Disease (CJD). Although no cases of
CJD have been directly linked to beef consumption, an advisory
committee has reported that 10 recent cases of a CJD variant may be
associated with BSE. This announcement has alarmed consumers well
beyond the borders of the United Kingdom.

Abstract: The incidence of bovine spongiform encephalopathy (BSE) in the
United Kingdom is now declining at a significant rate, indicating that
the 1988 ban on feeding ruminant-derived meat and bone meal to cattle
is having the anticipated effect. The question now is whether or not
BSE can be completely eradicated. At present there is no evidence of
lateral or maternal transmission, the occurrence of which would
complicate the eradication process. Eradication therefore seems to be
achievable, especially now that meat and bone meal has been recently
banned from the diets of all farmed animals in the UK. In this review
the aetiological role of meat and bone meal in the causation of BSE is
discussed together with the epidemiological data and the results of
studies on genetic susceptibility. The controversial theories relating
to the nature of the causal agent, and strain-typing studies on BSE
agent, are described. Current information on pathogenesis and diagnosis
is presented. The occurrence of BSE in cattle outside the United
Kingdom, and BSE-related disease in species other than cattle is also
discussed.

Abstract: Creutzfeldt-Jakob Disease (CJD), a neurodegenerative and
dementing disease of later life, is caused by a viruslike entity that
is incompletely characterized, As in scrapie, all more purified
infectious brain preparations contain nucleic acids. However, it has
not been possible to visualize unique bands that may derive from a
viral genome, We here used a subtractive strategy known as
representational difference analysis (RDA) to uncover such sequences.
To reduce the complexity of starting target nucleic acids, sucrose
gradients were first used to select nuclease resistant particles with a
defined 120S size, In CJD this single 120S gradient peak is highly
enriched for infectivity, and contains reduced amounts of PrP (Proc.
Natl. Acad. Sci, 92, 5124-8, 1995). Parallel 120S fractions from
uninfected brain were made to generate subtractor sequences. 120S
particles were lysed in GdnSCN, and ng amounts of released RNA were
purified for random-primed cDNA synthesis. To capture representative
fragments of 100 - 500 bp, cDNAs were cleaved with Mbo I for adaptor
ligation and amplification. In the first experiment with moderate RDA
selection, it was possible to Visualize clones from CJD cDNA that did
not hybridize to control cDNA, In the second experiment, more
exhaustive subtractions yielded a discrete set of CJD derived gel
bands. Competitive hgbridization showed a subset of these bands were
not present in either the control 120S cDNA or in the hamster genome,
This represents tile first demonstration of apparently CJD-specific:
nucleic acid bands in more purified infectious preparations. Although
exhaustive cloning, sequencing and correlative titration studies need
to be done, it is encouraging that most of the viral candidates
selected thus far have no significant homology with any previously
described sequence in the database.

Abstract: Converging advances in the fields of molecular biology, molecular
genetics, cellular biology and embryology have given researchers the
tools for the targeted delivery and stable germline transmission of
foreign genes (transgenes) in the mouse. In the realm of
neuropsychiatry, this so-named transgenic technology has begun to
catapult our knowledge of the genetic, molecular and cellular
mechanisms in the brain that are linked to a variety of mental
disorders as well as normal behavioral processes such as learning and
memory. Moreover, the potential for transgenic technology to develop
more precise animal models of specific human neuropsychiatric disorders
will likely lead to the identification of novel targets for therapeutic
intervention and facilitate the preclinical testing of new therapeutic
agents. This article examines key issues and highlights recent
developments pertaining to the transgenic modeling of neuropsychiatric
disorders.

Abstract: The appearance of bovine spongiform encephalopathy (BSE) followed
by new spongiform encephalopathies and variant Creuzfeldt-Jakob disease
(CJD) in the United Kingdom indicates that these diseases may be
linked. To give an understanding of this risk, the authors review the
literature on the pathogenesis of CJD and BSE and the current findings
on how these diseases are transmitted. They also discuss the
implications for Canada's food and blood supply and outline previously
published recommendations for disease prevention.

neurodegenerative diseases that includes Creutzfeldt-Jakob disease,
kuru, Gerstmann-Straussler syndrome, and fatal familial insomnia in
human beings. Also called prion diseases, these disorders are unique in
being both infectious and genetic in origin. Mutations in the human
gene that encodes the prion protein (PrP) are strongly linked to
inherited forms of the disease. The agent responsible for infectious
forms of the disease is thought to be a protein called PrPSc. prp(Sc)
cor responds to an altered form of the normal PrPC. Several pieces of
evidence indicate that PrPSc and PrPC do not differ in their primary
amino acid sequence, but rather in their conformations. Prion
replication,, is believed to occur when PrPSc in the infecting inoculum
interacts specifically with host PrPC, catalyzing its conversion into
PrPSc. Recent in vitro studies as well as the use of transgenic animals
have given new insights into the comprehension of the mechanisms of
these diseases. The conversion of PrPC into PrPSc is now considered as
the central event in the transmission as well as in the pathogenic
process of prion diseases.

Abstract: The report of the Creuhfeldt-Jakob Surveillance Unit from March
1996 regarding 10 cases of a new variant of Creutzfeldt-Jakob disease
(CJD) in young adults caused a great deal of uproar when it was
suggested that a possible Link with bovine spongiform encephalopathy
(BSE) could not be excluded. BSE was first noticed in 1986 after the
introduction of modified rendering systems in the manufacture of meat
and bone meal containing animal wastes contaminated with scrapie-like
agents. This article reviews available information on CSD, BSE and
other diseases caused by prions, transmission studies and the report of
the CJD Surveillance Unit and discuses possible links between BSE and
the new variant of CJD.

Abstract: The occurence of Creutzfeldt-Jakob disease (CJD) in two teenagers
and four dairy farmers in the United-Kingdom has focused attention on
the possibility of a causal link between bovine spongiform
encephalopathy (BSE) and Creutzfeldt-Jakob disease.
The BSE epidemic was initiated by the inclusion in the cattle feed
of ruminant-derived protein in 1981/1982. A feed ban of the
ruminant-derived protein in 1988 prevented further transmission of the
infective agent to cattle by this route. However cases in animals born
some years after the feeban suggest that there has been some continued
leakage of BSE infected material into animal feed (and perhaps maternal
transmission?).
To protect public and animal health, measures have been taken in
the United Kingdom, especially prohibition of specified bovine offals
in 1989 (extending this ban to thymus and intestine of calves
slaughtered for human consumption in 1994). These measures are
considered as minimizing the risk of BSE infection.
Although the BSE linked risk factors are probably unique to the
United Kingdom, it seems necessary to have the same regulations for the
calves imported from this country. More, it is crucial that the ongoing
surveillance programme of CJD in Europe is sustained.

Abstract: In the last 40 years, the pathogenesis of transmissible
spongiform encephalopathies of humans and animals have been the subject
of exciting discoveries and also of passionate controversies. Although
the human forms of these diseases are rare, the recent epidemics of
bovine spongiform encephalopathy has most dramatically raised the issue
of the transmissibility from affected animals to humans. This review
summarizes some current hypotheses on the nature of the infectious
agent causing these diseases, and on the pathogenetic pathways which
produce histologically detectable damage to the central nervous system.
The main focus is on the use of transgenic and knockout mice which,
combined with organ transplantation and bone marrow reconstitution
techniques, have provided powerful tools for dissecting the
pathogenesis of these diseases.
